    Spin-crossover, mesomorphic and thermoelectrical properties of cobalt(II) complexes with alkylated N3-Schiff bases

    Three new cobalt(ii) complexes, [Co(L12)2](BF4)2 (1), [Co(L14)2](BF4)2·H2O (2) and [Co(L16)2](BF4)2·H2O (3), where L12-16 are N3-Schiff bases appended with linear C12-16 carbon chains at the nitrogen atoms, were obtained in good yields by facile one-pot reactions. The single crystal X-ray structure of complex 1 shows a tetragonally compressed CoN6 coordination geometry. The melting temperatures of 1-3 were lower than 373 K, while their decomposition temperatures were above 473 K. All complexes have high-spin Co(ii) centres at 300 K and exhibit a columnar mesophase above 383 K. Complexes 1 and 3 showed normal thermal spin-crossover behaviour with weak hysteresis loops at about 320 K. Hence, these complexes showed uncoupled phase transitions (class iiia). The values for the Seebeck coefficient (Se) of the cobalt redox couples formed from 1 and 2 were 1.89 ± 0.02 mV K-1 and 1.92 ± 0.08 mV K-1, respectively, identifying them as potential thermoelectrochemical materials

    Estimation of genetic parameters for milk traits in Romanian local sheep breed

    ABSTRACT Objective. Estimate the genetic parameters for milk traits in a Romanian local sheep population Teleorman Black Head. Material and methods. Records of 262 sheep belonging to 17 rams and 139 ewes were used in the study. The following traits were investigated: milk yield, fat yield, protein yield, fat percentage and protein percentage. The genetic parameters were estimated using the Restricted Maximum Likelihood method, with a model including maternal effects. Results. The results from our study revealed that direct heritability estimates were moderate for milk yield (0.449), fat yield (0.442), protein yield (0.386) while for protein percentage (0.708) and fat percentage (0.924) were high. The high direct and maternal genetic correlation was between milk yield and protein yield (0.979, 0.973) and between protein yield and fat yield (0.952, 0.913) while the phenotypic correlation between the milk yield and fat yield (0.968), the milk yield and protein yield (0.967), fat yield and protein yield (0.936) was high and positive. Conclusions. The genetic parameters are important in selection program on this breed for genetic improvement

    The crystal structure of Trz1, the long form RNase Z from yeast.

    tRNAs are synthesized as precursor RNAs that have to undergo processing steps to become functional. Yeast Trz1 is a key endoribonuclease involved in the 3΄ maturation of tRNAs in all domains of life. It is a member of the β-lactamase family of RNases, characterized by an HxHxDH sequence motif involved in coordination of catalytic Zn-ions. The RNase Z family consists of two subfamilies: the short (250-400 residues) and the long forms (about double in size). Short form RNase Z enzymes act as homodimers: one subunit embraces tRNA with a protruding arm, while the other provides the catalytic site. The long form is thought to contain two fused β-lactamase domains within a single polypeptide. Only structures of short form RNase Z enzymes are known. Here we present the 3.1 Å crystal structure of the long-form Trz1 from Saccharomyces cerevisiae. Trz1 is organized into two β-lactamase domains connected by a long linker. The N-terminal domain has lost its catalytic residues, but retains the long flexible arm that is important for tRNA binding, while it is the other way around in the C-terminal domain. Trz1 likely evolved from a duplication and fusion of the gene encoding the monomeric short form RNase Z

    Relationship between gene polymorphism and milk production traits in Teleorman Black Head sheep breed

    ABSTRACT Objective. This study is a preliminary step of a larger national program aimed to develop a strategy for “in situ” preservation of Teleorman Black Head sheep population. In this paper we estimated the effect of β-lactoglobulin, casein and prolactin on some quantitative and qualitative milk traits in this local sheep population. Material and methods. Genotyping methodology included PCR for CSN3 (A and B alleles) and PCR-RFLP for LGB (A and B alleles) and PRL (T and C alleles), respectively. Repeated milking and milk composition analysis were used for the polymorphism effect estimation. Results. No association between CSN3 polymorphism and milk traits was found. Effect of LGB on production traits was quite constant. Genotype AA performed better than BB. PRL marker effect showed small differences than LGB. Concerning milk, fat and protein yield, AA genotype for PRL had a smaller positive impact than AA genotype for LGB. Regarding fat and protein content, PRL showed a negative effect for AA and positive for BB genotype, respectively. Conclusions. Positive association between LGB and milk yield and composition recommend this candidate gene like marker for a future MAS program. Although PRL gene is also associated with an increased milk quantity, inverse response over milk composition must be considered in MAS strategy. Our study demonstrated that both LGB and PRL markers could became an advent of MAS utilization in Romanian dairy sheep breeding industry.   RESUMEN Objetivo. Este estudio es un paso preliminar de un programa nacional más amplio destinado a desarrollar una estrategia para la conservación “in situ”de la población de ovejas Cabeza Negra de Teleorman. En este trabajo se estimó el efecto de la β-lactoglobulina, caseína y prolactina en algunos rasgos cuantitativos y cualitativos de la leche en esta población de ovejas locales. Material y métodos. Metodología de PCR para genotipificación incluido CSN3 (A y B alelos) y PCR-RFLP para LGB (A y B alelos) y PRL (T y C alelos). Análisis y composición de la leche de ordeños repetidos se utilizaron para estimación el efecto del polimorfismo. Resultados. No se encontró asociación entre el polimorfismo y la leche rasgos CSN3. Efecto de LGB en los rasgos de producción era bastante constante. Genotipo AA obtenido mejores resultados que BB. Efecto marcador PRL mostró pequeñas diferencias que LGB. En cuanto a la leche grasa y proteína el genotipo AA para PRL tuvo un impacto positivo más pequeño que el genotipo AA para LGB. En cuanto a contenido de grasa y proteína, PRL mostró un efecto negativo para AA y positivo para BB genotipo. Conclusiones.La asociación positiva entre LGB y la producción de leche y la composición recomienda este gen candidato como marcador para un futuro programa de MAS. Aunque gen PRL también se asocia con un incrementoen la cantidad de leche, la respuesta inversa sobre composición de la leche debe ser considerado en la estrategia de MAS. Nuestro estudio demostró que los marcadores tanto LGB y PRL podrían venir a ser utilizados en MAS en la industria rumanade cría deovejas lecheras

    Serbian Citizens’ Opinion on the COVID-19 Epidemic

    Aim: The Centre for International Public Policy has undertaken a public opinion research in which we tested the opinions of the citizens of Serbia on the coronavirus epidemic. The respondents had the opportunity to express their opinion on measures undertaken by the Serbian Government to combat the virus, to state their trust in the media, as well as the health system in general. In addition, we tested the prevalence of different conspiracy theories among citizens, whether the pandemic gave China a new image in the minds of the people and, most importantly, the level of solidarity among Serbian citizens as well as within the European / international community as a whole. Methods: In seven days, from 8-15th April 2020, we gathered a convenient sample of N=5989 respondents, which makes this the largest public opinion research project in Serbia on the topic of COVID-19 since the start of the epidemic. The electronic questionnaire consisted of 24 questions of mixed and closed type. Results: The findings of this research suggest that citizens of Serbia are not afraid of COVID-19, but are nevertheless cautious (86%). The percentage of those willing to consult a doctor when they notice any symptoms lies at 70%. Half of the respondents do not believe in alternative theories regarding the origin of the COVID-19 virus. The majority of the respondents (55%) hold government officials accountable for spreading panic through public speeches and daily public addresses. Moreover, 60% of the respondents do not trust the Serbian media outlets that are currently reporting on the COVID-19 pandemic. Furthermore, over half of the respondents are prepared to report their neighbour when he or she is coming from abroad and violates the obligation to self-isolate. However, mostly due to the significant fines, 65% of the respondents would not report the elderly when they are breaking the limited-movement restriction measures. Conclusion: As before the epidemic, opinions of the Serbian population on current topics are somewhat polarized. Although the majority of the respondents are cautious, a significant number also believes in conspiracy theories and does not fully trust the information provided by the media or the government.                                                                               &nbsp

    Fuzzy Fibers: Uncertainty in dMRI Tractography

    Fiber tracking based on diffusion weighted Magnetic Resonance Imaging (dMRI) allows for noninvasive reconstruction of fiber bundles in the human brain. In this chapter, we discuss sources of error and uncertainty in this technique, and review strategies that afford a more reliable interpretation of the results. This includes methods for computing and rendering probabilistic tractograms, which estimate precision in the face of measurement noise and artifacts. However, we also address aspects that have received less attention so far, such as model selection, partial voluming, and the impact of parameters, both in preprocessing and in fiber tracking itself. We conclude by giving impulses for future research

    Visible light photoactivity of Polypropylene coated Nano-TiO2 for dyes degradation in water

    The use of Polypropylene as support material for nano-TiO2 photocatalyst in the photodegradation of Alizarin Red S in water solutions under the action of visible light was investigated. The optimization of TiO2 pastes preparation using two commercial TiO2, Aeroxide P-25 and Anatase, was performed and a green low-cost dip-coating procedure was developed. Scanning electron microscopy, Atomic Force Microscopy and X-Ray Diffraction analysis were used in order to obtain morphological and structural information of as-prepared TiO2 on support material. Equilibrium and kinetics aspects in the adsorption and successive photodegradation of Alizarin Red S, as reference dye, are described using polypropylene-TiO2 films in the Visible/TiO2/water reactor showing efficient dyes degradation

    DEFECTIVE KERNEL1 regulates cellulose synthesis and affects primary cell wall mechanics

    The cell wall is one of the defining features of plants, controlling cell shape, regulating growth dynamics and hydraulic conductivity, as well as mediating plants interactions with both the external and internal environments. Here we report that a putative mechanosensitive Cys-protease DEFECTIVE KERNEL1 (DEK1) influences the mechanical properties of primary cell walls and regulation of cellulose synthesis. Our results indicate that DEK1 is an important regulator of cellulose synthesis in epidermal tissue of Arabidopsis thaliana cotyledons during early post-embryonic development. DEK1 is involved in regulation of cellulose synthase complexes (CSCs) by modifying their biosynthetic properties, possibly through interactions with various cellulose synthase regulatory proteins. Mechanical properties of the primary cell wall are altered in DEK1 modulated lines with DEK1 affecting both cell wall stiffness and the thickness of the cellulose microfibril bundles in epidermal cell walls of cotyledons

    Measurement batch differences and between-batch conversion of Alzheimer's disease cerebrospinal fluid biomarker values

    Introduction: Batch differences in cerebrospinal fluid (CSF) biomarker measurement can introduce bias into analyses for Alzheimer's disease studies. We evaluated and adjusted for batch differences using statistical methods. Methods: A total of 792 CSF samples from 528 participants were assayed in three batches for 12 biomarkers and 3 biomarker ratios. Batch differences were assessed using Bland-Altman plot, paired t test, Pitman-Morgan test, and linear regression. Generalized linear models were applied to convert CSF values between batches. Results: We found statistically significant batch differences for all biomarkers and ratios, except that neurofilament light was comparable between batches 1 and 2. The conversion models generally had high R2 except for converting P-tau between batches 1 and 3. Discussion: Between-batch conversion allows harmonized CSF values to be used in the same analysis. Such method may be applied to adjust for other sources of variability in measuring CSF or other types of biomarkers

    Factors Affecting the Folding of Pseudomonas aeruginosa OprF Porin into the One-Domain Open Conformer

    Pseudomonas aeruginosa OprF is a largely monomeric outer membrane protein that allows the slow, nonspecific transmembrane diffusion of solutes. This protein folds into two different conformers, with the majority conformer folding into a two-domain conformation that has no porin activity and the minority conformer into a one-domain conformation with high porin activity and presumably consisting of a large β barrel. We examined the factors that control the divergent folding pathways of OprF. OprF contains four Cys residues in the linker region connecting the N-terminal β-barrel domain and the C-terminal globular domain in the majority conformer. Prevention of disulfide bond formation either by expression of OprF in an Escherichia coli dsbA strain grown with dithiothreitol or by replacement of all Cys residues with serine (CS OprF) increased the specific pore-forming activity of OprF significantly. Replacement of Phe160 with Ile at the end of the β-barrel termination signal as well as replacement of Lys164 in the linker region with Gly, Cys, or Glu increased porin activity 2-fold. Improving a potential β-barrel termination signal in the periplasmic domain by replacement of Asp211 with asparagine also increased porin activity. The porin activity could be improved about 5-fold by the combination of these replacements. OprF mutants with higher porin activity were shown to contain more one-domain conformers by surface labeling of the A312C residue in intact cells, as this residue is located in the periplasmic domain in the two-domain conformers. Finally, when the OprF protein was expressed in an E. coli strain lacking the periplasmic chaperone Skp, the CS OprF protein exhibited increased pore-forming activity
